Combinib ( lapatinib 250 mg )

Combinib (Lapatinib 250 mg) is a generic kinase inhibitor that is used for the treatment of Breast Cancer. 

Lapatinib 250mg is indicated for the following:

1. Advanced or metastatic breast cancer patients whose tumors overexpress human epidermal growth factor receptor 2 (HER2) and who have received prior therapy, including an anthracycline, a taxane, and trastuzumab. Capecitabine is used for the treatment of such patients. It is important to note that patients should have experienced disease progression on trastuzumab before initiating treatment with lapatinib in combination with capecitabine.

2. Postmenopausal women with hormone receptor-positive metastatic breast cancer that overexpresses the HER2 receptor for whom hormonal therapy is indicated can be treated with Letrozole. lapatinib in combination with an aromatase inhibitor has not been compared to a trastuzumab-containing chemotherapy regimen for the treatment of metastatic breast cancer.

Lapatinib Dosage

To treat advanced or metastatic breast cancer, it is recommended to take Lapatinib orally once daily at a dose of 1,250 mg (equivalent to 5 tablets) on Days 1-21 continuously, in combination with capecitabine 2,000 mg/m2/day administered orally in 2 doses approximately 12 hours apart on Days 1-14, in a repeating 21-day cycle.

For hormone receptor-positive, HER2-positive metastatic breast cancer, Lapatinib should be taken orally once daily at a dose of 1,500 mg (equivalent to 6 tablets) continuously, in combination with letrozole at a dose of 2.5 mg once daily. It is recommended to take Lapatinib at least one hour before or one hour after a meal, but capecitabine should be taken with food or within 30 minutes after food. It is important to take Lapatinib as a single daily dose and not divide the dose.

The recommended dose of Lapatinib should be modified in case of cardiac and other toxicities, severe hepatic impairment, diarrhea, and CYP3A4 drug interactions.

Before and during treatment, your healthcare provider should monitor your heart function.

Diarrhea is a common side effect of Lapatinib and may sometimes be severe, leading to dehydration and even death. If you experience a change in bowel pattern or severe diarrhea, contact your healthcare provider immediately. They can provide instructions on how to prevent or treat diarrhea.

Lapatinib may also cause lung problems, such as a cough that does not go away or shortness of breath, as well as severe skin reactions, including skin rash, red skin, blistering of the lips, eyes, or mouth, peeling of the skin, and fever. If you experience any of these symptoms, notify your healthcare provider immediately. They may instruct you to stop taking Lapatinib.

Common side effects of Lapatinib in combination with capecitabine or letrozole include diarrhea, red and painful hands and feet, nausea, rash, vomiting, inflamed mouth, digestive tract, and airways, mouth sores, headache, unusual hair loss or thinning, shortness of breath, dry skin, itching, tiredness, painful arms, legs, and back, loss of appetite, indigestion, nosebleeds, and nail disorders such as nail bed changes, nail pain, infection, and swelling of the cuticles. If you experience any side effects that bother you or do not go away, inform your healthcare provider.
