Indications & Dosage

EVERMIL 5 (Everolimus 5mg) is a generic kinase inhibitor indicated for the treatment of:

1.Postmenopausal women with advanced hormonereceptor-positive,HER2- negative breast cancer in combination with exemestane after failure of treatment with letrozole or anastrozole.

2.Adults with progressive neuroendocrine tumors of pancreatic origin (PNET) and adults with progressive, well-differentiated, non-functional neuroendocrine tumors (NET) of gastrointestinal (GI) or lung origin that are unresectable, locally advanced or metastatic. Limitation of Use: AFINITOR is not indicated for the treatment of patients with functional carcinoid tumors.

3.Adults with advanced renal cell carcinoma (RCC) after failure of treatment with sunitinib or sorafenib.

4.Adults with renal angiomyolipoma and tuberous sclerosis complex (TSC), not requiring immediate surgery.

AFINITOR is a kinase inhibitors indicated for the treatment of adult and pediatric patients aged 1 year and older with TSC who have subependymal giant cell astrocytoma (SEGA) that requires therapeutic intervention but cannot be curatively resected

Everolimus Dosage

Do not combine AFINITOR to achieve the total daily dose.

Modify the dose for patients with hepatic impairment or for patients taking drugs that inhibit or induce P-glycoprotein (P-gp) and CYP3A4.

Breast Cancer:

1.10 mg orally once daily.

NET:  10 mg orally once daily.

RCC:  10 mg orally once daily.

TSC-Associated Renal Angiomyolipoma:

 10 mg orally once daily.

TSC-Associated SEGA: · 4.5 mg/m2 orally once daily; adjust dose to attain trough concentrations of 5-15 ng/mL.

Everolimus side effects

Lung or Breathing Problems: In some patients, lung or breathing problems may be severe and can even lead to death. Patients should tell their health care provider right away if they have any of these symptoms: new or worsening cough, shortness of breath, chest pain, difficulty breathing, or wheezing. 

Infections: Everolimus may make patients more likely to develop an infection, such as pneumonia, or a bacterial, fungal, or viral infection. Viral infections may include reactivation of hepatitis B in people who have had hepatitis B in the past. In some people, these infections may be severe and can even lead to death. Patients may need to be treated as soon as possible. Patients should tell their health care provider right away if they have a temperature of 100.5°F or above, have chills, or do not feel well. Symptoms of hepatitis B or infection may include the following: fever, chills, skin rash, joint pain and inflammation, tiredness, loss of appetite, nausea, pale stools or dark urine, yellowing of the skin, or pain in the upper right side of the stomach.

Severe Allergic Reactions: Severe allergic reactions can happen in people who take Everolimus. Call your health care provider or get medical help right away if you get signs and symptoms of a severe allergic reaction including: rash, itching, hives, flushing, trouble breathing or swallowing, chest pain or dizziness.

Angioedema: Patients who take an angiotensin-converting enzyme (ACE) inhibitor medicine during treatment with Everolimus are at a possible increased risk for a type of allergic reaction called angioedema. Talk with your health care provider before taking Everolimus if you are not sure if you take an ACE inhibitor medicine. Get medical help right away if you have trouble breathing or develop swelling of your tongue, mouth, or throat during treatment with Everolimus

Kidney Failure: Patients taking Everolimus may develop kidney failure. In some people, this may be severe and can even lead to death. Patients should have tests to check their kidney function before and during their treatment with Everolimus.

Delayed Wound Healing: Everolimus can cause incisions to heal slowly or not heal well. Call your health care provider right away if your incision is red, warm, or painful; if you have blood, fluid, or pus in your incision; if your incision opens up; or if your incision swells.

Increased Blood Sugar and Fat (cholesterol and triglycerides) Levels in Blood: Your health care provider should do blood tests to check your fasting blood sugar, cholesterol, and triglyceride levels in the blood before you start and during treatment with Everolimus.

Decreased Blood Cell Counts: Everolimus can cause you to have decreased red blood cells, white blood cells, and platelets. Your health care provider should do blood tests to check your blood cell counts before you start and during treatment with Everolimus.
